Downregulation of serum miR-26a predicts poor clinical outcome of papillary thyroid carcinoma
نویسندگان
چکیده
Deregulation of miRNAs has been demonstrated to play an important role in the initiation and development of many types of cancers including papillary thyroid carcinoma (PTC). Currently the role of miR-26a and its potential clinical significance for PTC remains unknown. Eighty-four PTC patients and forty healthy controls were enrolled in this study. Quantitative real-time PCR (qRT-PCR) was performed to determine the expression level of serum miR-26a in all the participants. The association between serum miR-26a expression level and clinical outcome of PTC was investigated. Serum miR-26a expression level was reduced in PTC patients compared with the healthy controls (P<0.01) and significantly increased after receiving treatments (P<0.01). Serum miR-26a expression level was associated with lymph node metastasis (P=0.0173) and TNM stage (P=0.0022). In addition, PTC with lower serum miR-26a expression had poorer overall (P=0.029) and disease free survival rates (P=0.0004). Moreover, serum miR-26a expression was an independent risk factor for PTC (P<0.05). Collectively, down-regulation of serum miR-26a is associated with poor prognosis in PTC and it might be a promising biomarker for this malignancy.

miR-26a and its Target CKS2 Modulate Cell Growth and Tumorigenesis of Papillary Thyroid Carcinoma
BACKGROUND While many studies have shown that levels of miR-26a are lower in papillary thyroid carcinoma (PTC), the role and mechanism of miR-26a in PTC are unclear. METHOD We used database searches to select potential mRNA targets of miR-26a. Anti-miR-26a, miR-26a mimic, siRNA for CKS2 and their effects on cell growth, cell-cycle distribution and colony formation were evaluated. We also eval...
متن کاملMicroRNAs (miRNAs) are small non-coding RNAs 18-25 nucleotides in length, transcribed from non-protein- coding genes or introns, which regulate gene expression through repressing translation and cleaving their target mRNAs by binding to complementary sites in their 3'-untrans-
Accumulating evidence indicates that microRNAs are involved in multiple processes in cancer development and progression. microRNA-26a (miR-26a) has been identified as a tumor suppressor and its downregulation is associated with poor prognosis in several types of human cancer. However, the specific function of miR-26a in osteosarcoma remains unclear. In the present study, we found that the expre...
متن کاملIL-6 upregulation contributes to the reduction of miR-26a expression in hepatocellular carcinoma cells
A recent study showed that miR-26a is downregulated in hepatocellular carcinoma tissues and that this downregulation is an independent predictor of survival. Interestingly, the same study also reported that miR-26a downregulation causes a concomitant elevation of IL-6 expression. Because miR-26a expression was found to be transcriptionally downregulated by oncogene c-Myc in various cancers, and...
متن کاملDown-regulation of miR-181b promotes apoptosis by targeting CYLD in thyroid papillary cancer.
MicroRNAs (miRNAs) are a small class of non-coding RNAs that are widely deregulated in various cancers. They act as either oncogenes or tumor suppressor genes in human cancer. The purpose of this study was to examine the potential role of miR-181b in human thyroid papillary cancer. The expression levels of different miRNAs were measured by micro array analysis in 10 thyroid papillary cancer spe...
متن کاملThyroid Papillary Microcarcinoma: Etiology, Clinical Manifestations, Diagnosis, Follow-up, Histopathology and Prognosis
Background: Thyroid carcinoma is the most common malignancy of the endocrine system. Papillary thyroid carcinoma (PTC) is the most common type of thyroid cancer, accounting for 70–90% of well-differentiated thyroid malignancies. Thyroid papillary microcarcinoma is a subtype of papillary carcinoma that included tumors with less than 10mm diameter. As a result of diagnostic methods improvement, p...
